I've run the next set of tires on wheels that were drilled with out putting the screws in. If you are running tubes I would recommend the screws be reinstalled but going tubeless I would put a shoe polish stripe on the tire at the valve stem to see if the tire is moving on the rim. If it is the screws might need to be reinstalled.
