I have just upgraded to a PT-54 and 50's. I liked tinkering with the fuel pressure on my old Thrasher chip setup.
Is it better to have the BLM lock at WOT or not?
 
You ~liked~ messing with the fuel pressure?!?! Since I've added a Trans+/Extender chip those days are gone.

But....to your question.

The WOT BLM lock (at 128) will make tuning at the track easier. It keeps the computer from compensating for your FP changes.

Driveability shouldn't be affected at all. In other words...it's a good idea.
