Setting Throttle Position Sensor?

What's the best method to dial in the TPS? When I set it for .42 at idle, the WOT numbers are lower than desired - 4.2 or 4.3. What's the trick? If the sensor is adjusted with the engine off, will the numbers remain unchanged once the engine is started? Will the additional voltage supplied by the alternator affect the TPS output?
